Transgenic and targeted gene modification analyses of zebrafishes and mice. (A and B) Effect of oca2 wt(A) or mt(G) mRNA overexpression in oca2 morphants. (A) The embryos at 36 h postfertilization were classified into four classes based on melanophore pigmentation. Class IV, wild-type pigmentation; Class I–III, different levels of pigmentation reduction. (B) Ratios of different classes of embryos. N, the total number of observed embryos. Effect of zebrafish oca2 knockdown on melanophore pigmentation. A:Test of oca2-MO effectiveness. Injection of poca2-EGFP plasmid DNA at the one-cell stage resulted gave rise to GFP during mid gastrulation, which was effectively blocked by co-injection of oca2-MO. B: Effect of oca2 knockdown on pigmentation. One-cell stage embryos were injected with 5 ng or 10 ng oca2-MO and observed for pigmentation at 36 or 48 hpf. The ratio of embryos with the representative pigmentation level was indicated. Note that embryos injected with 5 ng oca2-MO showed variable levels of pigment reduction in contrast to complete loss of pigmentation in embryos injected with 10 ng oca2-MO.
