The role of GPR171 in regulating embryonic HSC emergence is evolutionarily conserved in mice. (A) Limiting dilution transplantation analysis indicates the frequency of HSCs in the AGM and umbilical and vitelline arteries (A + U + V) of Gpr171+/+ and Gpr171−/− mouse embryos at embryonic day 11.5 (E11.5). (B) Western blotting analysis for protein levels of pERK1/2, total ERK1/2, and GAPDH in A + U + V cells of Gpr171+/+ and Gpr171−/− mouse embryos at E11.5. (C) Qualification of pERK1/2 and ERK1/2 signal density from (B) relative to GAPDH (n = 3; two-tailed, unpaired Student’s t test, *P < 0.05, ns, not significant. Data are presented as means ± SD). |
