The expression of igf2bp1 during early zebrafish embryo development. (A) Whole-mount in situ hybridization of igf2bp1 from 1-cell to 96hpf. Before 12 hpf and after 72 hpf, the expression of igf2bp1 is low, and igf2bp1 was mainly expressed in brain and liver of zebrafish embryos at 48 hpf and 72 hpf. (B) Igf2bp1 expression was detected by qRT-PCR during embryonic development in zebrafish.

Morpholino-mediated knockdown of igf2bp1. (A) Validation of knockdown efficiency of igf2bp1. Embryos were co-injected with Cont-MO and igf2bp1-GPF (a) or co-injected with igf2bp1-MO1 and igf2bp1-GPF (b). (B) In igf2bp1-MO2, splicing of exon 6 was disrupted with a 59 bp deletion and mRNA translation was prematurely terminated (b), which were indicated by the 633 bp RT-PCR products in Cont-MO and 574 bp in igf2bp1-MO2 (a). (C) No significant developmental malformation was detected in embryos of zebrafish. Cont-MO: Embryos injected with control MO. igf2bp1-MO1/2: Embryos injected with igf2bp1-MO1or igf2bp1-MO2.

Igf2bp1 is required for hepatic outgrowth but not for hepatoblast specification and budding. Whole-mount in situ hybridization of the early endoderm marker gene expression in Cont-MO (A, C, E and G) and igf2bp1-MO1/2 (B, D, F and H) embryos at 30 and 48 hpf.
