Expression of dlx1a in dlx2a−/− and expression of dlx2a in dlx1a−/− from 24hpf- 72hpf by WISH show little compensation except in 72hpf dlx1a−/−. Expression of dlx1a in dlx5a−/− and dlx6a−/− at 24hpf – 72hpf show little change in staining intensity compared to WT. Expression of dlx2a is increased in dlx5a−/− and dlx6a−/−at all stages, especially at 72hpf. Expression of dlx2a is present in the telencephalon, diencephalon and branchial arches. Expression of dlx2a is most increased in branchial arches in mutants whereas increases in dlx6a mutants are seen globally. Percentages indicate how often the phenotype was observed. Scale = 500 μm.

Expression of dlx5a in dlx1a−/− and dlx2a−/− is reduced to near undetectable levels by 72hpf. Expression of dlx5a is present in 24hpf – 72hpf WT and 24-48hpf mutants in the telencephalon, diencephalon, branchial arches, otic placode and pectoral fin buds. Percentages indicate how often the phenotype was observed. Scale = 500 μm.

Expression of dlx6a in dlx5a−/− and dlx5a in dlx6a−/− show dlx5a regulates dlx6a expression. Expression of dlx6a is reduced in the absence of dlx5a whereas the expression of dlx5a is increased in 48hpf and 72hpf dlx6a−/−, especially in the prethalamus (outline arrowhead) and hypothalamus (black arrowhead). Expression of dlx5a and dlx6a persists in 24hpf – 72hpf larvae despite deletion of intergenic enhancers in inter56−/− and is associated with increased dlx2a expression. The expression of dlx5a is lower than WT at 24hpf in inter56−/− but increases to levels comparable to WT by 48hpf. Expression of dlx6a is increased in all stages analyzed. Percentages indicate how often the phenotype was observed. Scale = 500 μm.
