Heat shock genes and the heat shock response in zebrafish embryos

Abstract
Heat shock genes exhibit complex patterns of spatial and temporal regulation during embryonic development in a wide range of organisms. Our laboratory has initiated an analysis of heat shock protein gene expression in the zebrafish, a model system that is now utilized extensively for the examination of early embryonic development of vertebrates. We have cloned members of the zebrafish hsp47, hsp70,\i and hsp90 gene families and shown them to be closely related to their counterparts in higher vertebrates. Whole mount in situ hybridization and Northern blot analyses have revealed that these genes are regulated in distinct spatial, temporal, and stress-specific manners. Furthermore, the tissue-specific expression patterns of the hsp47 and hsp90 alpha genes correlate closely with the expression of genes encoding known chaperone targets of Hsp47 and Hsp90 in other systems. The data raise a number of interesting questions regarding the function and regulation of these heat shock genes in zebrafish embryos during normal development and following exposure to environmental stress. Les modes de régulation spatio-temporelle des gènes du choc thermique au cours du développement embryonnaire sont complexes chez un large éventail d'organismes. Notre laboratoire a débuté une analyse de l'expression des gènes de protéines du choc thermique chez le poisson-zèbre, un système modèle souvent utilisé pour étudier le développement précoce d'embryons de vertébrés. Nous avons cloné des gènes des familles hsp47, hsp70 et hsp90 du poisson-zèbre et avons démontré qu'ils sont très apparentés à ceux des vertébrés supérieurs. L'hybridation in situ dans des montages d'élément entier et des transferts Northern ont révélé que ces gènes sont réglés de façon distinctive en fonction de l'espace, du temps et d'un stress. De plus, l'expression spécifique des gènes hsp47 et hsp90 alpha dans les tissus est fortement corrélée à l'expression de gènes codant des chaperonines cibles de Hsp47 et Hsp90 dans d'autres systèmes. Ces résultats soulèvent certaines questions intéressantes concernant la fonction et la régulation de ces gènes du choc thermique dans les embryons de poisson-zèbre durant le développement normal et à la suite d'une exposition à un stress environnemental.
